系统性红斑狼疮患者外周血单一核细胞CCR6、CCR8趋化因子受体mRNA的表达

| 目的 探讨趋化因子受体CCR6及CCR8在SLE患者外周血单个核细胞（PBMC）中的表达，与疾病的相关性。方法 收集93例确诊的SLE患者和30例正常对照，收集外周血单一核细胞，提取RNA。应用RT-PCR方法检测CCR6、CCR8 mRNA表达水平。记录患者临床表现及实验室检查。结果 1、CCR6 mRNA在PBMC中表达水平，活动期增高；CCR8 mRNA在PBMC中表达水平，活动期与非活动期及对照组之间差异无显著性。非活动期与对照组，患者组与对照组，两组间差异无显著性。2、SLE患者组CCR6 mRNA水平与疾病活动度评分关系：SLE患者组PBMC中CCR6 mRNA表达水平与SLEDAI分别作直线相关性分析，CCR6 mRNA水平与SLEDAI呈正相关。3、患者分为两组：具有某些临床表现的与不具临床表现的CCR6 mRNA及CCR8 mRNA表达，差异无统计学意义。结论 CCR6 mRNA表达水平在活动期SLE 比非活动期及对照组增高，与疾病的活动性（SLEDAI）正相关。CCR8 mRNA表达水平在SLE患者组与正常人对照组无显著性差异。CCR6参与疾病的发生及组织损伤。 |

| Objective We assessed the expression of chemokine receptors (CCR6 and CCR8) on peripheral blood mononuclear cell(PBMC) with active or inactive SLE and the correlation with SLE disease active index. Methods 93 definitive SLE patients and 30 healthy controls were collected. PBMC were separated from the peripheral blood specimen. Reverse transcription-polymerase chain reaction(RT-PCR) technique was applied to semi-quantitatively analyze the expression of chemokine receptors (CCR6 and CCR8) in PBMC. Results Level of CCR6 expression was significantly increased in patients with SLE as compared with controls, and significantly higher in patients with active SLE than those of the inactive and controls. Level of CCR6 expression in PBMC showed significantly positive correlation with disease activity(SLEDAI). Level of CCR8 expression was no diference between the actives and the inactives, the inactives and the controls. Conclusions These results suggest that CCR6 might be involved in the pathogenesis of SLE, and the expression level might be a good indicator for the disease activity of SLE. |
